Ipamorelin and CJC-1295 are among the most studied peptides in growth hormone regulation. Both are secretagogues, molecules that stimulate natural GH release in preclinical models.

Ipamorelin: selective GH secretagogue

Ipamorelin is a synthetic pentapeptide (711.9 Da), a selective ghrelin receptor (GHS-R) agonist. In preclinical models, it stimulates GH release without significant effects on cortisol, prolactin, or aldosterone. The effect depends on the dose, which allows precise titration in research protocols.

CJC-1295: GHRH analog

CJC-1295 with DAC is a modified peptide with a half-life of 6 to 8 days, producing longer-lasting GH and IGF-1 elevation. Mod GRF 1-29 (without DAC) has a shorter half-life of about 30 minutes and a more pulsatile GH release, closer to the natural pattern. Both available at Peptid.si.

Combination in research

CJC-1295 acts on the GHRH receptor (GH synthesis); Ipamorelin acts on the ghrelin receptor (pulsatile release). Preclinical models suggest that combining both pathways produces greater GH release than either peptide alone.

Limitations of evidence

Most data is preclinical. Human data exists for CJC-1295 with DAC from early clinical phases, but the studies never progressed to approval. Neither peptide is approved for medical use, and both remain research compounds.
